Ure(k)a! Sirtuins Regulate Mitochondria
نویسندگان
چکیده
Increasing evidence suggests that multiple metabolic pathways are regulated by sirtuin-dependent protein deacetylation in the mitochondria. In this issue, Nakagawa et al. (2009) show that the sirtuin SIRT5 deacetylates and activates a mitochondrial enzyme, carbamoyl phosphate synthetase 1, which mediates the first step in the urea cycle.

The sirtuins, oxidative stress and aging: an emerging link
Reactive oxygen species (ROS) are a family of compounds that can oxidatively damage cellular macromolecules and may influence lifespan. Sirtuins are a conserved family of nicotinamide adenine dinucleotide (NAD+)-dependent protein deacetylases that regulate lifespan in many model organisms including yeast and mice. Recent work suggests that sirtuins can modulate ROS levels notably during a dieta...
متن کاملRegulation of metabolism by sirtuins
Mitochondrial sirtuins are NAD-dependent enzymes that bind and regulate numerous metabolic pathways within the mitochondria. For example, SIRT3 functions as an NAD-dependent deacetylase that binds and activates numerous oxidative pathways. We have discovered that sirtuins regulate metabolic pathways important in tumor cell metabolism. One hallmark feature of tumor cells is a shift from oxidativ...
متن کاملMitochondrial protein acylation and intermediary metabolism: regulation by sirtuins and implications for metabolic disease.
The sirtuins are a family of NAD(+)-dependent protein deacetylases that regulate cell survival, metabolism, and longevity. Three sirtuins, SIRT3-5, localize to mitochondria. Expression of SIRT3 is selectively activated during fasting and calorie restriction. SIRT3 regulates the acetylation level and enzymatic activity of key metabolic enzymes, such as acetyl-CoA synthetase, long-chain acyl-CoA ...
متن کاملUrea cycle regulation by mitochondrial sirtuin, SIRT5
Mammalian sirtuins have diverse roles in aging, metabolism and disease. Recently we reported a new function for SIRT5 in urea cycle regulation. Our study uncovered that SIRT5 localized to mitochondria matrix and deacetylates carbamoyl phosphate synthetase 1 (CPS1), an enzyme which is the first and rate-limiting step of urea cycle. Deacetylation of CPS1 by SIRT5 resulted in activation of CPS1 en...
متن کاملSirtuins and Disease: The Road Ahead
Sirtuins represent a promising new class of conserved histone deacetylases, originally identified in yeast. The activity of the sirtuin (SirT) family - made up of seven members (SirT1-7) - is NAD(+) dependent. Sirtuins target a wide range of cellular proteins in nucleus, cytoplasm, and mitochondria for post-translational modification by acetylation (SirT1, 2, 3, and 5) or ADP-ribosylation (SirT...
